Product Overview

This rabbit polyclonal antibody recognizes mineralocorticoid receptor (gene NR3C2) in human, mouse and rat samples and is validated for ICC/IF and Western blot. The predicted molecular weight is 107.1 kDa, and the supplier reports an observed band at about 110 kDa.

The mineralocorticoid receptor (MR, NR3C2) is a nuclear receptor activated by aldosterone and also by cortisol. In kidney epithelia it drives genes that increase sodium and water retention and potassium excretion, raising blood pressure. MR is studied in blood pressure control, heart and kidney disease, and it is the target of spironolactone.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)
Immunocytochemistry / Immunofluorescence 5 µg/mL (Human)

Samples with a confirmed band (WB): human HeLa cells, human 293T cells, human LNCAP cells, human MCF-7 cells, rat NRK cells, mouse NIH/3T3 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (ICC/IF): U2OS cells.

Immunogen

Synthetic peptide from the C-terminal region of human NR3C2. It differs from the mouse sequence at one residue and from the rat sequence at two residues.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places mineralocorticoid receptor in the cytoplasm, nucleus and endoplasmic reticulum membrane. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat); Immunocytochemistry / Immunofluorescence 5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 107.1 kDa; observed band at about 110 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human NR3C2. It differs from the mouse sequence at one residue and from the rat sequence at two residues.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
