Product Overview
This rabbit polyclonal antibody recognizes mitotic interactor and substrate of PLK1 (gene MISP) in human samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 75.4 kDa, and the supplier reports an observed band at about 75 kDa.
MISP is an actin-binding protein phosphorylated by the mitotic kinase PLK1. It helps position the mitotic spindle by regulating dynactin at the cell cortex and stabilizing astral microtubule attachments, and it may also link microtubules to actin and focal adhesions during cell migration.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human MCF-7 cells, human HepG2 cells, human A549 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): Caco-2 cells.
Immunogen
Recombinant human MISP fragment (Met1–Arg632), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places mitotic interactor and substrate of PLK1 in the cell junction and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
