Product Overview
Clone 26M37 is a rabbit monoclonal antibody that recognizes mixed lineage kinase domain-like protein (gene MLKL) in human, mouse and rat samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 54.5 kDa, and the supplier reports an observed band at about 54 kDa.
MLKL (mixed lineage kinase domain-like) is a pseudokinase that carries out necroptosis, a regulated form of necrotic cell death. After phosphorylation by RIPK3, MLKL oligomerizes and moves to the plasma membrane, where it disrupts membrane integrity, and phospho-MLKL is commonly used as a marker of necroptosis in studies of inflammation and infection.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunoprecipitation | 1:50 |
Samples with a confirmed band (WB): human HeLa cells, human PC-3 cells, human A549 cells, human MCF-7 cells, rat testis tissue, rat liver tissue, mouse testis tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide based on the sequence of MLKL.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places mixed lineage kinase domain-like protein in the cytoplasm, cell membrane and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
