Product Overview

Clone CDG-13 is a rabbit monoclonal antibody that recognizes interstitial collagenase (gene MMP1) in human samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 54.0 kDa, and the supplier reports an observed band at about 54 kDa.

Interstitial collagenase (MMP-1) is a secreted zinc-dependent matrix metalloproteinase that cuts the triple helix of fibrillar collagens types I, II and III at a single site. This first cut starts collagen breakdown, so MMP-1 is studied in wound healing, arthritis, skin aging and tumor invasion.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry 1:50

Samples with confirmed staining (IHC): mouse kidney tissue, rat kidney tissue, mouse lung tissue, human astrocytoma tissue, human colon tissue, human hepatocellar carcinoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human MMP1.

Reactivity Notes

The supplier lists reactivity with human. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 54.0 kDa; observed band at about 54 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human MMP1.
