Product Overview

This rabbit polyclonal antibody recognizes collagenase 3 (gene Mmp13) in human, mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 53.4 kDa, and the supplier reports an observed band at about 54 kDa.

Collagenase 3 (MMP-13) is a matrix metalloproteinase that degrades fibrillar collagens, with the highest activity toward type II collagen of cartilage, and it also cleaves aggrecan and other matrix proteins. It is needed for normal bone development and is widely studied in cartilage breakdown in osteoarthritis.

Validated Applications

Western blot 0.1–0.5 µg/mL
IHC (paraffin sections) 0.5–1 µg/mL
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human A549 cells, human PC-3 cells, human U2OS cells, human HEK293 cells, rat testicular tissue, mouse testicular tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): mouse lung tissue, mouse small intestine tissue, rat lung tissue, rat small intestine tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Immunogen

Recombinant rat MMP13 fragment (Tyr99–His335), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt describes collagenase 3 as a secreted protein. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
