Product Overview
This rabbit polyclonal antibody recognizes neutrophil collagenase (gene Mmp8) in mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 53.1 kDa; the supplier reports observed bands at 55, 70 kDa.
Neutrophil collagenase (MMP-8) is a matrix metalloproteinase stored in neutrophil granules and released during inflammation. It cleaves fibrillar collagens types I, II and III and is studied in inflammatory tissue destruction, including periodontal disease.

Validated Applications
| IHC (paraffin sections) | 0.5–1 µg/mL (Mouse, Rat) |
|---|---|
| ELISA | 0.1–0.5 µg/mL |
| Western blot | 0.1–0.5 µg/mL (Mouse, Rat) |
Samples with a confirmed band (WB): rat spleen tissue, mouse spleen tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse spleen tissue, rat spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the N-terminal region of mouse MMP-8. It differs from the human sequence at eleven residues and from the rat sequence at nine residues.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places neutrophil collagenase in the cytoplasmic granule. Tissue expression noted by UniProt (mouse): Neutrophils. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
