Product Overview

Clone EFA-13 is a rabbit monoclonal antibody that recognizes neutrophil collagenase (gene MMP8) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 53.4 kDa, and the supplier reports an observed band at about 70 kDa.

Neutrophil collagenase (MMP-8) is a matrix metalloproteinase stored in neutrophil granules and released during inflammation. It cleaves fibrillar collagens types I, II and III and is studied in inflammatory tissue destruction, including periodontal disease.

Target Neutrophil collagenase (gene MMP8; UniProt P22894, human)

Samples with a confirmed band (WB): human placenta.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human spleen tissue, mouse spleen tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human MMP8.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places neutrophil collagenase in the cytoplasmic granule. Tissue expression noted by UniProt (human): Neutrophils. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 53.4 kDa; observed band at about 70 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human MMP8.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
