Product Overview
This rabbit polyclonal antibody recognizes ribosomal protein S6 kinase alpha-5 (gene RPS6KA5) in human, mouse and rat samples and is validated for ICC/IF, IHC and Western blot. The predicted molecular weight is 89.9 kDa, and the supplier reports an observed band at about 90 kDa.
MSK1 (RPS6KA5) is a mitogen- and stress-activated kinase activated by ERK and p38 MAPK. In the nucleus it phosphorylates CREB, ATF1, RelA and histones H3 and HMGN1, linking MAPK signaling to immediate-early gene induction, and it helps control inflammatory responses, for example by inducing IL-10 and DUSP1.

|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 0.5–1 µg/mL (Human) |

Samples with a confirmed band (WB): human HEL cells, rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse cardiac muscle tissue, rat brain tissue, human placenta tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): T47D cells.
Immunogen
Recombinant human MSK1 fragment (Val540–Gln665), expressed in E. coli. Sequence identity with the mouse ortholog: 98%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ribosomal protein S6 kinase alpha-5 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Widely expressed with high levels in heart, brain and placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
