Product Overview

Clone FHI-13 is a rabbit monoclonal antibody that recognizes cytochrome c oxidase subunit 2 (gene MT-CO2) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 25.6 kDa, and the supplier reports an observed band at about 21 kDa.

Cytochrome c oxidase subunit 2 (MT-CO2, COX2) is a mitochondrial DNA-encoded core subunit of complex IV of the respiratory chain. It contains the copper A center that accepts electrons from cytochrome c and passes them to subunit 1, where oxygen is reduced to water.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:20
Flow cytometry 1:50

Samples with a confirmed band (WB): K562 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human liver tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human MTCO2.

Reactivity Notes

The supplier lists reactivity with human. UniProt places cytochrome c oxidase subunit 2 in the mitochondrion inner membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on porcine samples?
A.Reactivity with porcine samples has not been tested by the supplier. Aligning the immunogen sequence with the porcine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 25.6 kDa; observed band at about 21 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human MTCO2.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
