Product Overview

Clone 22N23 is a rabbit monoclonal antibody that recognizes oxidized purine nucleoside triphosphate hydrolase (gene NUDT1) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 22.5 kDa, and the supplier reports an observed band at about 18 kDa.

MTH1 (NUDT1) is a Nudix hydrolase that cleans the nucleotide pool by breaking down oxidized purine nucleotides such as 8-oxo-dGTP and 2-oxo-dATP. This prevents their incorporation into DNA and the resulting mutations, and MTH1 is studied in oxidative stress, DNA damage and cancer cell survival.

Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:50
Flow cytometry 1:100

Samples with a confirmed band (WB): human Jurkat cells, human U2OS cells, human MCF-7 cells, human A549 cells, human THP-1 cells, human HeLa cells, human 293T cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human MTH1.

Reactivity Notes

The supplier lists reactivity with human. Tissue expression noted by UniProt (human): Widely expressed with highest expression in thymus, testis, embryo and proliferating blood lymphocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:100; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 22.5 kDa; observed band at about 18 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human MTH1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
