Product Overview
This rabbit polyclonal antibody detects mucin-1 (gene MUC1) in human samples and is validated for Western blot, IHC, ICC/IF and ELISA. The predicted molecular weight is 122.1 kDa, and the supplier reports an observed band at about 25–30 kDa.
Mucin-1 (MUC1) is a heavily glycosylated transmembrane mucin on the apical surface of most epithelia, forming part of the protective barrier. Its cytoplasmic tail engages signaling pathways including ERK, SRC and NF-κB and can influence p53-regulated transcription. MUC1 is often overexpressed and aberrantly glycosylated in carcinomas, so it is widely studied in cancer biology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human T47D cells, human HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human invasive adenocarcinoma of the lung tissue, human endometrioid adenocarcinom tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): human lung adenocarcinoma tissue.
Immunogen
Recombinant human MUC1 fragment (Thr1063–Arg1131), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places mucin-1 in the apical cell membrane. Tissue expression noted by UniProt (human): Expressed on the apical surface of epithelial cells, especially of airway passages, breast and uterus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
