Product Overview

Clone 27M55 is a rabbit monoclonal antibody that recognizes methylmalonyl-CoA mutase, mitochondrial (gene MMUT) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 83.1 kDa, and the supplier reports an observed band at about 83 kDa.

Methylmalonyl-CoA mutase (MMUT) is a mitochondrial enzyme that uses adenosylcobalamin, an active form of vitamin B12, to convert methylmalonyl-CoA into succinyl-CoA. This reaction feeds products of branched-chain amino acid and odd-chain fatty acid breakdown into the TCA cycle, and MMUT deficiency causes methylmalonic acidemia.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human K562 cells, human 293T cells, human PC-3 cells, rat PC-12 cells, rat kidney tissue, mouse NIH/3T3 cells, mouse kidney tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human MUTA.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places methylmalonyl-CoA mutase, mitochondrial in the mitochondrion matrix, mitochondrion and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
