Product Overview

Clone AFAC-13 is a rabbit monoclonal antibody that recognizes myosin-11 (gene MYH11) in human and mouse samples and is validated for Western blot and IHC. The predicted molecular weight is 227.3 kDa, and the supplier reports an observed band at about 250 kDa.

Smooth muscle myosin heavy chain (MYH11) is the motor protein of smooth muscle thick filaments, converting ATP hydrolysis into contraction. It is a specific marker of mature smooth muscle cells, and MYH11 mutations are linked to familial thoracic aortic aneurysm, while a CBFB-MYH11 fusion occurs in a subtype of acute myeloid leukemia.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200

Samples with a confirmed band (WB): human bladder.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human myosin, smooth muscle heavy chain 1 and 2.

Reactivity Notes

The supplier lists reactivity with human and mouse. UniProt places myosin-11 in the melanosome. Tissue expression noted by UniProt (human): Smooth muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 227.3 kDa; observed band at about 250 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human myosin, smooth muscle heavy chain 1 and 2.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
