Product Overview
This rabbit polyclonal antibody detects myoglobin (gene MB) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 17.2 kDa, and the supplier reports an observed band at about 17 kDa.
Myoglobin (MB) is a small, heme-containing oxygen-binding protein found in the cytoplasm of cardiac and skeletal muscle. It stores oxygen and helps it diffuse to mitochondria during muscle activity. Myoglobin released from damaged muscle appears in blood and urine, so it is widely measured in studies of muscle injury and cardiac damage.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse, Rat) |
Samples with a confirmed band (WB): rat heart tissue, rat skeletal muscle tissue, mouse heart tissue, mouse skeletal muscle tissue, rat liver tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse skeletal muscle tissue, rat skeletal muscle tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the N-terminal region of human myoglobin. It differs from the mouse sequence at three residues and from the rat sequence at six residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places myoglobin in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
