Product Overview
This rabbit polyclonal antibody targets serine/threonine-protein kinase TBK1 in human, mouse and rat samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 83.6 kDa, and the supplier reports an observed band at about 84 kDa.
TBK1 is a serine/threonine kinase that drives type I interferon responses to viral and bacterial infection. Recruited by adaptors such as STING, MAVS and TRIF, it phosphorylates IRF3 and IRF7 so they can enter the nucleus and induce interferon genes, and it also regulates selective autophagy; TBK1 variants are linked to ALS and frontotemporal dementia.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human 293T cells, human HepG2 cells, human LNCAP cells, rat brain tissue, rat C6 cells, mouse brain tissue, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse testis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human NAK/TBK1 fragment (Met1–Leu729), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places serine/threonine-protein kinase TBK1 in the cytoplasm. Tissue expression noted by UniProt (human): Ubiquitous with higher expression in testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
