Product Overview

This rabbit polyclonal antibody recognizes serine/threonine-protein kinase TBK1 in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 83.6 kDa, and the supplier reports an observed band at about 100 kDa.

TBK1 is a serine/threonine kinase that drives type I interferon responses to viral and bacterial infection. Recruited by adaptors such as STING, MAVS and TRIF, it phosphorylates IRF3 and IRF7 so they can enter the nucleus and induce interferon genes, and it also regulates selective autophagy; TBK1 variants are linked to ALS and frontotemporal dementia.

Target Serine/threonine-protein kinase TBK1 (gene TBK1; UniProt Q9UHD2, human)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG
Reactivity Human, Mouse, Rat

Formulation Per vial: 0.9 mg NaCl; 0.2 mg Na2HPO4; 0.05 mg thimerosal; 0.05 mg NaN3; plus stabilizers
Calculated MW 83.6 kDa
Observed MW 100 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat, Mouse)
Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse)

Samples with a confirmed band (WB): HeLa cells, rat testis tissue, rat liver tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): rat testis tissue, human prostatic cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide from the C-terminal region of human NAK. The peptide sequence is conserved in rat and mouse.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places serine/threonine-protein kinase TBK1 in the cytoplasm. Tissue expression noted by UniProt (human): Ubiquitous with higher expression in testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse); IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat, Mouse). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on porcine samples?
A.Reactivity with porcine samples has not been tested by the supplier. Aligning the immunogen sequence with the porcine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 83.6 kDa; observed band at about 100 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human NAK. The peptide sequence is conserved in rat and mouse.
