Biological background

Protein function (datasheet): Transcription regulator involved in inner cell mass and embryonic stem (ES) cells proliferation and self-renewal. Imposes pluripotency on ES cells and prevents their differentiation towards extraembryonic endoderm and trophectoderm lineages. Blocks bone morphogenetic protein-induced mesoderm differentiation of ES cells by physically interacting with SMAD1 and interfering with the recruitment of coactivators to the active SMAD transcriptional complexes. Acts as a transcriptional activator or repressor. Binds optimally to the DNA consensus sequence 5'-TAAT[GT][GT]-3' or 5'- [CG][GA][CG]C[GC]ATTAN[GC]-3'. Able to autorepress its expression in differentiating (ES) cells: binds to its own promoter following interaction with ZNF281/ZFP281, leading to recruitment of the NuRD complex and subsequent repression of expression. When overexpressed, promotes cells to enter into S phase and proliferation. .

Cellular localization (datasheet): Nucleus .

Tissue details (datasheet): Expressed in testicular carcinoma and derived germ cell tumors (at protein level). Expressed in fetal gonads, ovary and testis. Also expressed in ovary teratocarcinoma cell line and testicular embryonic carcinoma. Not expressed in many somatic organs and oocytes. .

Research relevance and current trends

  • Commonly studied in contexts related to Cancer,Cell Cycle,Cell Differentiation,2339,Developmental Biology,Embryogenesis,Embryonic Stem Cells,Epigenetics and Nuclear Signaling,Intracellular,Nuclear Signaling Pathways,Stem Cells,Surface Molecules,Transcription,Transcription Factors.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

I was wanting to use your anti-Nanog Rabbit Monoclonal antibody for IF for human embryonic stem cell on frozen tissues, but I want to know if it has been tested for this particular application. Has this antibody been tested and is this antibody a good choice for human embryonic stem cell identification?
It shows on the product datasheet, M00153 anti-Nanog Rabbit Monoclonal antibody has been validated for IF, WB on human tissues. We have an innovator award program that if you test this antibody and show it works in human embryonic stem cell in IHC-frozen, you can get your next antibody for free.
We have seen staining in human cerebellar vermis. Are there any suggestions? Is anti-Nanog Rabbit Monoclonal antibody supposed to stain cerebellar vermis positively?
Based on literature cerebellar vermis does express NANOG. Based on Uniprot.org, NANOG is expressed in cerebellar vermis, embryonic stem cell, teratocarcinoma, among other tissues. Regarding which tissues have NANOG expression, here are a few articles citing expression in various tissues: Embryonic stem cell, Pubmed ID: 12787504, 14990856, 16391521 Teratocarcinoma, Pubmed ID: 14702039
