Target SCN5A

Overview

Anti-NaV1.5 (SCN5A) (extracellular) Antibody-APC is an antibody targeting Sodium channel protein type 5 subunit alpha, Voltage-gated sodium channel subunit alpha Nav1.5, Long QT Syndrome 3, LQT3 Polyclonal raised in Rabbit (Allophycocyanin (APC)). This antibody is commonly used in FC, LCI to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Sodium channel protein type 5 subunit alpha, Voltage-gated sodium channel subunit alpha Nav1.5, Long QT Syndrome 3, LQT3 (also reported as Sodium channel protein type 5 subunit alpha, Voltage-gated sodium channel subunit alpha Nav1.5, Long QT Syndrome 3, LQT3).
  • Immunogen/epitope region: Extracellular, 11th loop (3rd loop of 3rd repeat)..
  • Homology note: Rat – identical, Human – 14 out of 16 residues identical. (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #ASC-045), and direct flow cytometry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: Allophycocyanin (APC) (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

Voltage-gated Na+ channels (NaV) are responsible for myocardial conduction and maintenance of the cardiac rhythm and are essential for the generation of action potentials and cell excitability.1 Dysfunction or disregulation of cardiac sodium channels can cause several disorders, including cardiac arrhythmias. The majority of Na+ channels in the mammalian heart are Tetrodotoxin (TTX)-insensitive NaV1.5.2 The putative structure of NaV1.5 consists of four homologous domains (I-IV), each containing six transmembrane segments (S1-S6). Mutations in the C-terminus of NaV1.5 were described in connection to Long QT syndrome and Brugada syndrome.1-2 Recent data have demonstrated selective expression of NaV1.5 in the mouse central nervous system and implicated a role for NaV1.5 in the physiology of the central nervous system.1

Research relevance and current trends

  • Mapping receptor/channel localization across neuronal subtypes and subcellular compartments.
  • Linking trafficking or surface expression changes to activity-dependent signaling and plasticity.
  • Using KO/KD or blocking-peptide concepts to strengthen antibody-based target assignment.

Common research applications

  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.
  • Live cell imaging (LCI): support extracellular-epitope detection on non-permeabilized cells when appropriate.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-APC.

Application abbreviations: CBE- Cell-based ELISA, FC- Flow cytometry, ICC- Immunocytochemistry, IE- Indirect ELISA, IF- Immunofluorescence, IFC- Indirect flow cytometry, IHC- Immunohistochemistry, IP- Immunoprecipitation, LCI- Live cell imaging, N- Neutralization, WB- Western blot. Species abbreviations: H- Human, M- Mouse, R- Rat.
