Product Overview
This rabbit polyclonal antibody detects nuclear cap-binding protein subunit 1 (gene NCBP1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 91.8 kDa, and the supplier reports an observed band at about 92 kDa.
NCBP1 (CBP80) is the large subunit of the nuclear cap-binding complex, which binds the 5′ cap of newly made RNA polymerase II transcripts together with NCBP2. The complex supports pre-mRNA splicing, 3′ end processing, mRNA export, nonsense-mediated decay and the pioneer round of translation.

Samples with a confirmed band (WB): human HeLa cells, human Jurkat cells, human HepG2 cells, human MCF-7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human ovarian cancer tissue, human lung cancer tissue, human colorectal adenocarcinoma tissue, human liver cancer tissue, human lung adenocarcinoma tissue, human esophageal squamous carcinoma tissue, mouse brain cerebral cortex tissue, mouse brain hippocampus tissue, rat brain hippocampus tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Recombinant human NCBP1 fragment (Tyr121–Ala790), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places nuclear cap-binding protein subunit 1 in the nucleus and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
