Product Overview
This rabbit polyclonal antibody targets nck-associated protein 1 (gene NCKAP1) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 128.8 kDa, and the supplier reports an observed band at about 129 kDa.
NCKAP1 (NAP1) is a core subunit of the WAVE regulatory complex, which activates the Arp2/3 complex downstream of the small GTPase RAC1. This drives branched actin assembly at the leading edge and the formation of lamellipodia, so NCKAP1 is studied in cell migration, neuronal development and endosomal signaling of BDNF through its receptor TrkB.

Samples with a confirmed band (WB): human HeLa cells, human 293T cells, human RT4 cells, human A549 cells, rat brain tissue, rat L6 cells, mouse brain tissue, mouse NIH3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Recombinant human NCKAP1 fragment (Ser2–Lys201), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 98.5% and 98%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places nck-associated protein 1 in the cell membrane and cell projection. Tissue expression noted by UniProt (human): Expressed in all tissues examined except peripheral blood leukocytes, with highest expression in brain, heart, and skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
