Product Overview

This rabbit polyclonal antibody targets nebulette (gene NEBL) in human, mouse and rat samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 116.5 kDa; the supplier reports observed bands at 35, 126 kDa.

Nebulette (NEBL) is a smaller, heart-enriched relative of nebulin. It binds actin at the Z-disk of cardiac sarcomeres, supports Z-disk assembly and may connect thin filaments to desmin intermediate filaments, so it is studied in cardiac muscle structure.

Target Nebulette (gene NEBL; UniProt O76041, human)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG
Reactivity Human, Mouse, Rat
Form Lyophilized
Formulation Per vial: 4 mg trehalose; 0.9 mg NaCl; 0.2 mg Na2HPO4
Calculated MW 116.5 kDa
Observed MW 35, 126 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.1–0.25 µg/mL (Human, Mouse, Rat)
IHC (paraffin sections) 2–5 µg/mL (Mouse, Rat)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human SH-SY5Y cells, human MCF-7 cells, mouse lung tissue, rat heart tissue, mouse heart tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): mouse heart tissue, rat heart tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Recombinant human NEBL fragment (Glu86–Lys842), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Abundantly expressed in cardiac muscle, but not in skeletal or smooth muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.25 µg/mL (Human, Mouse, Rat); IHC (paraffin sections) 2–5 µg/mL (Mouse, Rat); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 116.5 kDa; observed bands at about 35 and 126 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant human NEBL fragment (Glu86–Lys842), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
