Product Overview
This rabbit polyclonal antibody detects E3 ubiquitin-protein ligase NEDD4 in human and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 149.1 kDa, and the supplier reports an observed band at about 115 kDa.
NEDD4 is a HECT-type E3 ubiquitin ligase with WW domains that recognize PY motifs in its substrates. It tags membrane receptors, channels and signaling proteins, such as IGF1R and ENaC, for endocytosis and lysosomal degradation, and it is studied in growth factor signaling, ion transport and virus budding.

Samples with a confirmed band (WB): human U251 cells, human RT4 cells, human Hacat cells, human A549- WT cells, human A549-NEDD4 KO cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat brain tissue, human colon adenocarcinoma tissue, human liver cancer tissue, human lung cancer tissue, human renal cells carcinoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): U87 cells.

Recombinant human NEDD4 fragment (Met1–Ala736), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and rat. UniProt places E3 ubiquitin-protein ligase NEDD4 in the cytoplasm, nucleus and cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
