Product Overview
This rabbit polyclonal antibody recognizes ubiquitin-like protein NEDD8 in human, mouse and rat samples and is validated for flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 9.1 kDa, and the supplier reports an observed band at about 12 kDa.
NEDD8 is a ubiquitin-like protein attached to a small set of targets, most importantly the cullin scaffolds of cullin-RING E3 ubiquitin ligases. This neddylation activates the ligases and drives degradation of cyclins and other regulators, so NEDD8 is central to cell cycle control, and the pathway is studied with inhibitors such as MLN4924 (pevonedistat).

Samples with a confirmed band (WB): human HepG2 cells, human MCF-7 cells, human RT4 cells, human PC-3 cells, rat brain tissue, rat thymus tissue, mouse brain tissue, mouse thymus tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): A431 cells.
Immunogen
Synthetic peptide from the middle region of human NEDD8. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ubiquitin-like protein NEDD8 in the nucleus. Tissue expression noted by UniProt (human): Highly expressed in heart, skeletal muscle, spleen, thymus, prostate, testis, ovary, colon and leukocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
