Product Overview
Clone AOAE-14 is a rabbit monoclonal antibody that recognizes ubiquitin-like protein NEDD8 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 9.1 kDa, and the supplier reports an observed band at about 10 kDa.
NEDD8 is a ubiquitin-like protein attached to a small set of targets, most importantly the cullin scaffolds of cullin-RING E3 ubiquitin ligases. This neddylation activates the ligases and drives degradation of cyclins and other regulators, so NEDD8 is central to cell cycle control, and the pathway is studied with inhibitors such as MLN4924 (pevonedistat).

Samples with a confirmed band (WB): human HepG2 cells, human MCF-7 cells, human RT4 cells, human PC-3 cells, rat brain tissue, rat heart tissue, mouse brain tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ubiquitin-like protein NEDD8 in the nucleus. Tissue expression noted by UniProt (human): Highly expressed in heart, skeletal muscle, spleen, thymus, prostate, testis, ovary, colon and leukocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
