Anti-β1-Adrenergic Receptor (extracellular) Antibody (#AAR-023) is a highly specific antibody directed against an epitope of the rat protein. The antibody can be used in western blot, immunohistochemistry and indirect live cell flow cytometry applications. It has been designed to recognize β1-Adrenergic Receptor from rat, mouse, and human samples. Anti-β1-Adrenergic Receptor (extracellular)- mFluor™ Violet 450 Antibody (#AAR-023-V) is directly conjugated to mFluor™ Violet 450 fluorophore. This conjugated antibody has been developed to be used in immunofluorescent applications such as direct flow cytometry and live cell imaging. mFluor™ Violet 450 dye has a maximum absorption of 405 nm and maximum emission of 445 nm. These spectral characteristics make it an excellent replacement for Pacific Blue™ or Brilliant Violet™ 421 dyes. For flow cytometry applications, the labeled antibody can be detected using the 405 nm laser lane and a filter set of 450/50.

Overview

Anti-β1-Adrenergic Receptor (extracellular)- mFluor™ Violet 450 Antibody is an antibody targeting Beta-1 adrenoceptor, Beta-1 adrenoreceptor, ADRB1, ADRB1R, B1AR Polyclonal raised in Rabbit (mFluor™ Violet 450. Maximum absorption 405 nm and maximum emission 445 nm. These spectral characteristics make it an excellent replacement for Pacific Blue™ or Brilliant Violet™ 421 dyes. For flow cytometry applications, the labeled antibody can be detected using the 405 nm laser lane and a filter set of 450/50.). This antibody is commonly used in FC, LCI to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Beta-1 adrenoceptor, Beta-1 adrenoreceptor, ADRB1, ADRB1R, B1AR (also reported as Beta-1 adrenoceptor, Beta-1 adrenoreceptor, ADRB1, ADRB1R, B1AR).
  • Homology note: Rat, Mouse, human - 14/15 amino acid residues identical. (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #AAR-023), and direct flow cytometry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: mFluor™ Violet 450. Maximum absorption 405 nm and maximum emission 445 nm. These spectral characteristics make it an excellent replacement for Pacific Blue™ or Brilliant Violet™ 421 dyes. For flow cytometry applications, the labeled antibody can be detected using the 405 nm laser lane and a filter set of 450/50. (may affect detection channel and background).
  • Antibody type: Polyclonal / Rabbit IgG.

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

β-Adrenergic receptors (βAR) belong to the superfamily of G-protein coupled receptors (GPCRs). βAR stimulation by the sympathetic nervous system or circulating catecholamines is involved in peripheral blood circulation, metabolic regulation, muscle contraction, and central neural activities. In the heart, acute βAR stimulation serves as the most powerful means to regulate cardiac output in response to a fight-or-flight situation.

Research relevance and current trends

  • Mapping receptor/channel localization across neuronal subtypes and subcellular compartments.
  • Linking trafficking or surface expression changes to activity-dependent signaling and plasticity.
  • Using KO/KD or blocking-peptide concepts to strengthen antibody-based target assignment.

Common research applications

  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.
  • Live cell imaging (LCI): support extracellular-epitope detection on non-permeabilized cells when appropriate.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-APC.

Application abbreviations: CBE- Cell-based ELISA, FC- Flow cytometry, ICC- Immunocytochemistry, IE- Indirect ELISA, IF- Immunofluorescence, IFC- Indirect flow cytometry, IHC- Immunohistochemistry, IP- Immunoprecipitation, LCI- Live cell imaging, N- Neutralization, WB- Western blot.
