Product Overview
Clone FBH-14 is a rabbit monoclonal antibody that recognizes nestin in human, mouse, rat and pig samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 177.4 kDa, and the supplier reports an observed band at about 270–300 kDa.
Nestin (NES) is a class VI intermediate filament protein expressed mainly in dividing neural progenitor cells and other stem or progenitor cells. It helps reorganize vimentin filaments during mitosis and is required for progenitor survival and self-renewal, and it is one of the most widely used markers of neural stem and progenitor cells.

Samples with a confirmed band (WB): human U251 cells, human SH-SY5Y cells, rat C6 cells, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human diffuse large B-cells lymphoma of the intestine tissue, human glioma tissue, human non-small cells lung cancer tissue, mouse brain tissue, rat brain tissue, pig brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): human brain tissue, rat brain tissue.
Immunogen
Synthetic peptide based on the sequence of human nestin.
Reactivity Notes
The supplier lists reactivity with human, mouse, rat and pig. Tissue expression noted by UniProt (human): CNS stem cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
