Product Overview
Clone AFBH-20 is a rabbit monoclonal antibody that recognizes substance-P receptor (gene TACR1) in human, mouse and rat samples and is validated for immunoprecipitation, IHC and Western blot. The predicted molecular weight is 46.3 kDa, and the supplier reports an observed band at about 48 kDa.
The substance P receptor (TACR1, also called neurokinin 1 receptor or NK1R) is a G protein-coupled receptor with highest affinity for substance P among the tachykinins. It signals mainly through phospholipase C and Ca2+ release, and it is studied in pain, neurogenic inflammation, nausea and emesis.

|---|---|
| Immunohistochemistry | 1:50–1:200 |
| Immunoprecipitation | 1:50 |
Samples with a confirmed band (WB): human Raji cells, human K562 cells, human MOLT-4 cells, rat spleen tissue, mouse kidney tissue, mouse spleen tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, mouse colon tissue, rat colon tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human neurokinin 1 Receptor.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places substance-P receptor in the cell membrane and early endosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
