Overview

This antibody is intended for detection of Elane (Neutrophil elastase) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-Neutrophil Elastase/ELANE Antibody Picoband® catalog # PB10058. Tested in ELISA, IHC, WB applications. This antibody reacts with Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E. coli-derived mouse Elastase/ELANE/ELA2 recombinant protein (Position: S27-N265). Mouse Elastase/ELANE/ELA2 shares 92% amino acid (aa) sequence identity with rat Elastase/ELANE/ELA2. (reported region: S27-N265).
  • Molecular weight context: reported MW: 29 kDa; calculated MW: 29 kDa
  • Reactivity: Mouse,Rat
  • Applications: ELISA, IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Neutrophil elastase; Neutrophil elastase. Neutrophil elastase is a serine protease of neutrophil and monocyte granules. Its key physiologic role is in innate host defense, but it can also participate in tissue remodeling and possesses secretagogue actions important to local inflammatory responses. Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode structurally similar proteins. The encoded preproprotein is proteolytically processed to generate the active protease. Following activation, this protease hydrolyzes proteins within specialized neutrophil lysosomes, called azurophil granules, as well as proteins of the extracellular matrix. This protein also degrades the outer membrane protein A (OmpA) of E. coli as well as the virulence factors of such bacteria as Shigella, Salmonella and Yersinia. Mutations in this gene are associated with cyclic neutropenia and severe congenital neutropenia (SCN). This gene is present in a gene cluster on chromosome 19. Functional note: Medullasin modifies the functions of natural killer cells, monocytes and granulocytes. Inhibits C5a-dependent neutrophil enzyme release and chemotaxis (By similarity). . Reported localization: Nucleus. Expression/tissue context: Found in all tissue examined including heart, brain, placenta, lung, liver, skeletal muscle, kidney and pancreas.

Research relevance and current trends

  • Calcium Signaling: Researchers commonly examine how Elane (Neutrophil elastase) relates to this theme using model systems and orthogonal readouts.
  • Cancer: Researchers commonly examine how Elane (Neutrophil elastase) relates to this theme using model systems and orthogonal readouts.
  • Complement: Researchers commonly examine how Elane (Neutrophil elastase) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative Elane (Neutrophil elastase) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of Elane (Neutrophil elastase) across tissue regions and cell types using matched controls.
  • ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
