Rabbit Monoclonal

Product Overview

Clone AOCI-5 is a rabbit monoclonal antibody that recognizes neutrophil elastase (gene ELANE) in human samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 28.5 kDa, and the supplier reports an observed band at about 29 kDa.

Neutrophil elastase (ELANE) is a serine protease stored in the azurophil granules of neutrophils. It degrades bacterial proteins and extracellular matrix components such as elastin, and it is studied in host defense, lung injury and inflammatory disease. ELANE mutations cause severe congenital and cyclic neutropenia.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human neutrophil Elastase.

Reactivity Notes

The supplier lists reactivity with human. UniProt places neutrophil elastase in the cytoplasmic vesicle, lysosome, cytolytic granule and nucleus. Tissue expression noted by UniProt (human): Bone marrow cells.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
