Product Overview
This rabbit polyclonal antibody recognizes merlin (gene NF2) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 69.7 kDa, and the supplier reports an observed band at about 69 kDa.
Merlin (NF2) is a membrane-cytoskeleton linker protein of the ERM family that acts as a tumor suppressor. It helps activate the Hippo pathway kinases LATS1/2 and mediates contact inhibition of growth, and loss of NF2 causes neurofibromatosis type 2 with schwannomas and meningiomas.

Samples with a confirmed band (WB): human HepG2 cells, human Caco-2 cells, human U251 cells, rat kidney tissue, rat C6 cells, mouse brain tissue, mouse HEPA1–6 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human mammary cancer tissue, human intestinal cancer tissue, mouse lung tissue, rat intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): A549 cells, HL-60 cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
