Product Overview
This rabbit polyclonal antibody detects nuclear factor of activated T-cells 5 (gene NFAT5) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 165.8 kDa, and the supplier reports an observed band at about 220 kDa.
NFAT5 (TonEBP) is a Rel family transcription factor that, unlike other NFAT proteins, is not controlled by calcineurin. It is the main regulator of the cellular response to high osmolarity, switching on genes that protect cells from hypertonic stress, and it also influences inflammatory gene expression.

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human Jurkat cells, human U2OS cells, human SiHa cells, human RT4 cells, rat PC-12 cells, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human bladder cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with confirmed staining (ICC/IF): human bladder cancer tissue.
Samples with a confirmed signal (flow cytometry): HeLa cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places nuclear factor of activated T-cells 5 in the nucleus, cytoplasm and chromosome. Tissue expression noted by UniProt (human): Widely expressed, with highest levels in skeletal muscle, brain, heart and peripheral blood leukocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
