Product Overview
This rabbit polyclonal antibody recognizes nuclear factor 1 A-type (gene NFIA) in human, mouse and rat samples and is validated for flow cytometry, IHC and Western blot. The predicted molecular weight is 55.9 kDa, and the supplier reports an observed band at about 56–70 kDa.
NFIA is a member of the nuclear factor I family of transcription factors, which binds the palindromic DNA motif TTGGC(N5)GCCAA in cellular and viral promoters, where it can activate transcription, and also supports adenovirus DNA replication. NFIA is important in brain development, particularly for the switch from neurogenesis to gliogenesis and astrocyte formation.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse, Rat) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human MCF-7 cells, rat liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): MCF-7 cells.
Immunogen
Synthetic peptide from the middle region of human NFIA. The peptide sequence is conserved in rat. It differs from the mouse sequence at one residue.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places nuclear factor 1 A-type in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
