Product Overview

This rabbit polyclonal antibody targets beta-nerve growth factor (gene NGF) in human samples and is validated for Western blot. The predicted molecular weight is 27.0 kDa, and the supplier reports an observed band at about 27 kDa.

Nerve growth factor (NGF) is the founding member of the neurotrophin family. Acting through its high-affinity receptor TrkA (NTRK1) and p75NTR (NGFR), it supports survival and growth of sympathetic and sensory neurons, while its precursor proNGF can signal cell death through p75NTR and sortilin family co-receptors; NGF is also a major mediator of pain.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human)

Samples with a confirmed band (WB): human U-87MG cells, human 22RV1 cells, human A549 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Immunogen

Synthetic peptide from the C-terminal region of human NGF. Sequence identity with the mouse and rat orthologs: 91.3%.

Reactivity Notes

The supplier lists reactivity with human. UniProt describes beta-nerve growth factor as secreted and also located in the endosome lumen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
