Product Overview
This rabbit polyclonal antibody targets nitric oxide synthase 1 (gene NOS1) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 161.0 kDa, and the supplier reports an observed band at about 160 kDa.
Neuronal nitric oxide synthase (nNOS, NOS1) is a Ca2+/calmodulin-dependent enzyme that converts L-arginine to nitric oxide. In the brain and peripheral nerves, nitric oxide acts as an unconventional neurotransmitter, and nNOS is also expressed in skeletal muscle. It is studied in synaptic plasticity, neurotoxicity and neuromuscular physiology, and it can promote S-nitrosylation of target proteins.

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Immunogen
Synthetic peptide from the C-terminal region of human nNOS(neuronal). The peptide sequence is conserved in mouse. It differs from the rat sequence at one residue.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places nitric oxide synthase 1 in the cell membrane and cell projection. Tissue expression noted by UniProt (human): Isoform 1 is ubiquitously expressed: detected in skeletal muscle and brain, also in testis, lung and kidney, and at low levels in heart, adrenal gland and retina. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
