Vendor description (summary): Boster Bio Anti-NNT Antibody Picoband® catalog # A00887-3.

Biological background

Biological context: Catalyzes the initial step in utilization of glucose by the beta-cell and liver at physiological glucose concentration. Glucokinase has a high Km for glucose, and so it is effective only when glucose is abundant. The role of GCK is to provide G6P for the synthesis of glycogen. Pancreatic glucokinase plays an important role in modulating insulin secretion. Hepatic glucokinase helps to facilitate the uptake and conversion of glucose by acting as an insulin-sensitive determinant of hepatic glucose usage.

Expression and localization notes: cellular localization: Cytoplasm., tissue context: Isoform 1 is expressed in pancreas. Isoform 2 and isoform 3 is expressed in liver..

Common research applications

  • Western blotting (WB): Compare NNT levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of NNT in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify NNT-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Background: This gene encodes an integral protein of the inner mitochondrial membrane. The enzyme couples hydride transfer between NAD (H) and NADP (+) to proton translocation across the inner mitochondrial membrane. Under most physiological conditions, the enzyme uses energy from the mitochondrial proton gradient to produce high concentrations of NADPH. The resulting NADPH is used for biosynthesis and in free radical detoxification.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cytoplasm.
  • Tissue details: Isoform 1 is expressed in pancreas. Isoform 2 and isoform 3 is expressed in liver.
  • Research category: Cancer,Cancer Metabolism,Carbohydrate Metabolism,Energy Metabolism,Energy Transfer Pathways,Metabolic Signaling Pathway,Metabolic Signaling Pathways,Metabolism,Metabolism Of Carbohydrates,Pathways and Processes,Signal Transduction
