Product Overview
This rabbit polyclonal antibody recognizes reticulon-4 (gene RTN4) in human samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 129.9 kDa, and the supplier reports an observed band at about 180 kDa.
Reticulon-4 (RTN4), also known as Nogo, is a membrane protein that shapes the tubular endoplasmic reticulum. Its long isoform, Nogo-A, is abundant in CNS myelin and inhibits axon growth through receptors such as NgR1, making it a major subject of research on limited axon regeneration after spinal cord and brain injury.

Samples with a confirmed band (WB): human U-87MG cells, human SH-SY5Y cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human prostatic cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with confirmed staining (ICC/IF): human prostatic cancer tissue.
Samples with a confirmed signal (flow cytometry): SiHa cells.
