Product Overview
Clone 22R65 is a rabbit monoclonal antibody that recognizes reticulon-4 receptor (gene RTN4R) in human, mouse and rat samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 50.7 kDa, and the supplier reports an observed band at about 70 kDa.
The Nogo receptor NgR1 (RTN4R) is a GPI-anchored neuronal surface protein that recognizes several myelin-associated growth inhibitors, including Nogo-A, MAG and OMgp. Working with co-receptors such as p75NTR and LINGO1, it switches on RhoA signaling that remodels the actin cytoskeleton and halts axon growth, so it is widely studied in spinal cord injury, axon regeneration and synaptic plasticity.

Validated Applications
| Western blot | 1:1000–1:5000 |
|---|---|
| Immunoprecipitation | 1:50 |
Samples with a confirmed band (WB): human HepG2 cells, human U2OS cells, human SH-SY5Y cells, human U251 cells, rat brain tissue, rat C6 cells, mouse brain tissue, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human nogo Receptor.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places reticulon-4 receptor in the cell membrane, membrane raft, cell projection and perikaryon. Tissue expression noted by UniProt (human): Widespread in the brain but highest levels in the gray matter. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
