Product Overview
This rabbit polyclonal antibody recognizes 28S rRNA (gene NOP2) in human samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 89.3 kDa, and the supplier reports an observed band at about 120 kDa.
NOP2 (also called NSUN1 or p120) is a nucleolar S-adenosylmethionine-dependent methyltransferase that places 5-methylcytosine at a specific site in 28S rRNA. It is also needed for pre-rRNA processing and 60S subunit biogenesis, partly through non-catalytic interactions with box C/D snoRNAs. It is studied in ribosome biogenesis and cell proliferation.

Samples with a confirmed band (WB): human HEL cells, human Caco-2 cells, human A549 cells, human HeLa cells, human RT4 cells, human 293T tissue, human SIHA cells lyates, human U251 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): RT4 cells.

Recombinant human NOP2 fragment (Leu210–Gln747), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places 28S rRNA in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
