Product Overview

This rabbit polyclonal antibody targets neurogenic locus notch homolog protein 2 (gene NOTCH2) in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 265.4 kDa; the supplier reports observed bands at 110 kDa, 300 kDa.

NOTCH2 is a Notch family receptor that is activated by Jagged and Delta-like ligands, leading to release of its intracellular domain and RBPJ-dependent gene activation. It regulates cell fate in tissues including liver, kidney, bone and B cells. NOTCH2 mutations underlie Alagille syndrome type 2 and Hajdu–Cheney syndrome.

Validated Applications

Western blot 0.25–0.5 µg/mL
IHC (paraffin sections) 2–5 µg/mL
Flow cytometry (fixed cells) 1–3 µg/1×106 cells
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human U87 cells, human MCF-7 cells, human 293T cells, human SH-SY5Y cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human glioblastoma tissue, human lung cancer tissu, mouse brain tissue, rat brain tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Samples with a confirmed signal (flow cytometry): SH-SY5Y cells.

Immunogen

Recombinant human NOTCH2 fragment (His2194–Ala2471), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places neurogenic locus notch homolog protein 2 in the cytoplasm. Tissue expression noted by UniProt (human): Expressed in the brain, heart, kidney, lung, skeletal muscle and liver. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL; IHC (paraffin sections) 2–5 µg/mL; Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells; ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 265.4 kDa; observed bands at about 110 and 300 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human NOTCH2 fragment (His2194–Ala2471), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
