Product Overview
This rabbit polyclonal antibody recognizes cytosolic 5'-nucleotidase 1A (gene NT5C1A) in human samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 41.0 kDa, and the supplier reports an observed band at about 41 kDa.
Cytosolic 5′-nucleotidase 1A (NT5C1A) hydrolyzes nucleoside monophosphates, mainly AMP, to release nucleosides such as adenosine and inorganic phosphate. It is highly expressed in skeletal muscle and is studied in adenosine production and energy metabolism, and autoantibodies against it are found in inclusion body myositis.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human SH-SY5Y cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human skeletal muscle tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human NT5C1A fragment (Glu2–Gln368), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places cytosolic 5'-nucleotidase 1A in the cytoplasm. Tissue expression noted by UniProt (human): Highly expressed in skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
