Product Overview

This rabbit polyclonal antibody targets nuclear respiratory factor 1 (gene NRF1) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 53.5 kDa, and the supplier reports an observed band at about 75 kDa.

Nuclear respiratory factor 1 (NRF1) is a transcription factor that activates many nuclear genes needed for mitochondrial respiration and heme synthesis, as well as TFAM and other factors that copy and transcribe mitochondrial DNA. It works with PGC-1 coactivators in mitochondrial biogenesis and is studied in energy metabolism and cell growth. It is distinct from NFE2L1, also called NRF1.

Target Nuclear respiratory factor 1 (gene NRF1; UniProt Q16656, human)

Validated Applications

Immunocytochemistry / Immunofluorescence 0.5–1 µg/mL (Human, Mouse, Rat)
IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat, Mouse)
Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse)

Samples with a confirmed band (WB): U2OS cells, HeLa cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human mammary cancer tissue, rat intestine tissue, human intestinal cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Samples with confirmed staining (ICC/IF): A549 cells.

Immunogen

Synthetic peptide from the middle region of human NRF1. The peptide sequence is conserved in rat and mouse.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places nuclear respiratory factor 1 in the nucleus. Tissue expression noted by UniProt (human): Ubiquitously expressed with strongest expression in skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse); IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat, Mouse); Immunocytochemistry / Immunofluorescence 0.5–1 µg/mL (Human, Mouse, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 53.5 kDa; observed band at about 75 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the middle region of human NRF1. The peptide sequence is conserved in rat and mouse.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
