Product Overview

Clone AEBF-14 is a rabbit monoclonal antibody that recognizes nuclear mitotic apparatus protein 1 (gene NUMA1) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 238.3 kDa, and the supplier reports an observed band at about 270 kDa.

NuMA (NUMA1) is a large microtubule-binding protein that is nuclear in interphase and moves to the spindle poles in mitosis. It tethers microtubule minus ends at the poles and works with dynein and LGN to orient the spindle, so it is studied in cell division, asymmetric division and nuclear organization.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human HeLa cells, human SH-SY5Y cells, human Hacat cells, human MCF-7 cells, human HEK293- WT cells, human HEK293-NUMA1 KO cells, rat testis tissue, rat brain tissue, mouse testis tissue, mouse brain tissueIHC:human bladder cancer tissue, human breast cancer tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human nuMA.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places nuclear mitotic apparatus protein 1 in the nucleus, nucleus matrix, chromosome and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on horse samples?
A.Reactivity with horse samples has not been tested by the supplier. Aligning the immunogen sequence with the horse ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 238.3 kDa; observed band at about 270 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human nuMA.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
