Rabbit Monoclonal

Product Overview

Clone 26N26 is a rabbit monoclonal antibody that recognizes nuclear pore glycoprotein p62 (gene NUP62) in human, mouse and rat samples and is validated for ICC/IF and Western blot. The predicted molecular weight is 53.3 kDa, and the supplier reports an observed band at about 70 kDa.

Nup62 is an O-GlcNAc-modified FG-repeat nucleoporin at the central channel of the nuclear pore complex, where it is central to nucleocytoplasmic transport. Outside the pore, it also associates with centrosomes and contributes to centrosome segregation and spindle orientation during mitosis.

Validated Applications

Western blot 1:1000–1:8000
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human PC-3 cells, human HeLa cells, human 293T cells, human Jurkat cells, rat testis tissue, rat RH35 cells, mouse testis tissue, mouse NIH/3T3 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (ICC/IF): HeLa cells.

Immunogen

Synthetic peptide based on the sequence of human NUP62.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places nuclear pore glycoprotein p62 in the nucleus, cytoplasm and nucleus envelope. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:8000; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 53.3 kDa; observed band at about 70 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human NUP62.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
