Product Overview
Clone 19P20 is a rabbit monoclonal antibody that recognizes POU domain, class 2, transcription factor 1 (gene POU2F1) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 76.5 kDa, and the supplier reports an observed band at about 90 kDa.
OCT1 (POU2F1) is a widely expressed POU-domain transcription factor that binds the octamer motif ATTTGCAT. It activates genes such as histone H2B, some small nuclear RNAs and immunoglobulins, and can modulate steroid receptor signaling. OCT1 also helps herpes simplex virus activate its immediate-early genes, and it is studied in stress and metabolic responses.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
Samples with a confirmed band (WB): Hhuman 293T cells, human K562 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, human stomach cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human 43374.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places POU domain, class 2, transcription factor 1 in the nucleus. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
