Product Overview
This rabbit polyclonal antibody detects origin recognition complex subunit 1 (gene ORC1) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 97.4 kDa, and the supplier reports an observed band at about 100 kDa.
ORC1 is the largest subunit of the origin recognition complex (ORC), which binds DNA replication origins in an ATP-dependent manner. ORC recruits CDC6, CDT1 and the MCM helicase to form pre-replication complexes that license origins for DNA replication, and ORC1 mutations cause Meier–Gorlin syndrome.

Samples with a confirmed band (WB): human HL-60 cells, human K562 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): U2OS cells.
Immunogen
Recombinant human ORC1 fragment (His628–Glu861), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places origin recognition complex subunit 1 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
