Product Overview
This rabbit polyclonal antibody targets ATP-dependent translocase ABCB1 in human, mouse and rat samples and is validated for flow cytometry, ICC/IF and Western blot. The predicted molecular weight is 141.5 kDa, and the supplier reports an observed band at about 160 kDa.
P-glycoprotein (ABCB1, MDR1) is an ATP-powered efflux pump in the plasma membrane that expels a wide range of drugs and moves certain phospholipids across the membrane. High ABCB1 levels are a classic cause of multidrug resistance in cancer cells, and it also shapes drug absorption and the blood-brain barrier.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| Immunocytochemistry / Immunofluorescence | 2 µg/mL |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells |
Samples with a confirmed band (WB): human THP-1 cells, human A375 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): U2OS cells.
Immunogen
Synthetic peptide from the middle region of human P Glycoprotein. Sequence identity with the rat ortholog: 62.9%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ATP-dependent translocase ABCB1 in the cell membrane, apical cell membrane and cytoplasm. Tissue expression noted by UniProt (human): Expressed in small intestine. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
