Overview
Anti-P2Y12/P2ry12 Antibody Picoband® is an antibody for P2ry12 detection raised in Rabbit (Polyclonal, Rabbit IgG), with reported reactivity: Mouse,Rat. Commonly used in WB, IHC, Flow Cytometry, ELISA workflows.
Key elements and design rationale
- Target: P2ry12 (Alpha-galactosidase A); UniProt: Q9CPV9
- Antibody format: Rabbit, Polyclonal, Rabbit IgG
- Molecular weight: 40 kDa, calculated 47643 MW
- Applications: WB, IHC, Flow Cytometry, ELISA
Vendor description (summary): Boster Bio Anti-P2Y12/P2ry12 Antibody Picoband® catalog # A01136-1.
Biological background
Biological context: Factor D cleaves factor B when the latter is complexed with factor C3b, activating the C3bbb complex, which then becomes the C3 convertase of the alternate pathway. Its function is homologous to that of C1s in the classical pathway.
Expression and localization notes: cellular localization: Lysosome..
Common research applications
- Western blotting (WB): Compare P2ry12 levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of P2ry12 in tissue sections, considering fixation and antigen retrieval effects.
- Flow cytometry: Quantify P2ry12-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Background: P2Y12 is a chemoreceptor for adenosine diphosphate (ADP) that belongs to the Gi class of a group of G protein-coupled (GPCR) purinergic receptors. The product of this gene belongs to the family of G-protein coupled receptors. This family has several receptor subtypes with different pharmacological selectivity, which overlaps in some cases, for various adenosine and uridine nucleotides. This receptor is involved in platelet aggregation, and is a potential target for the treatment of thromboembolisms and other clotting disorders. Mutations in this gene are implicated in bleeding disorder, platelet type 8 (BDPLT8). Alternative splicing results in multiple transcript variants of this gene.
- Cross reactivity: No cross-reactivity with other proteins.
- Cellular localization: Lysosome.
- Research category: Mesenchymal Stem Cells,Stem Cells
