Product Overview

Clone DEO-20 is a rabbit monoclonal antibody that recognizes cellular tumor antigen p53 (gene TP53) in human, mouse and rat samples and is validated for Western blot, ICC/IF and immunoprecipitation. The predicted molecular weight is 43.7 kDa, and the supplier reports an observed band at about 55 kDa.

p53 (TP53) is a tumor suppressor transcription factor that responds to DNA damage and other stresses by activating genes for cell cycle arrest, DNA repair, senescence or apoptosis. It is mutated in about half of human cancers, making it one of the most studied proteins in cancer biology, with rat models widely used alongside human cells.

Validated Applications

Western blot 1:500–1:2000
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:20

Immunogen

Synthetic peptide based on the sequence of human p53 (acetyl K370).

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places cellular tumor antigen p53 in the cytoplasm, nucleus, endoplasmic reticulum and mitochondrion matrix. Tissue expression noted by UniProt (human): Ubiquitous.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunocytochemistry / Immunofluorescence 1:50–1:200; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 43.7 kDa; observed band at about 55 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human p53 (acetyl K370).
