Product Overview

Clone EHI-20 is a rabbit monoclonal antibody that recognizes tumor protein p73 (gene TP73) in human samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 69.6 kDa, and the supplier reports an observed band at about 73 kDa.

p73 (TP73) is a transcription factor of the p53 family. Isoforms with a transactivation domain can promote apoptosis after DNA damage, whereas ΔN isoforms lacking this domain oppose p53 and full-length p73. p73 is also needed for the development of multiciliated cells and parts of the nervous system, and it is studied in cancer and neurodevelopment.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human THP-1 cells, human U2OS cells, human Caco-2 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Samples with confirmed staining (IHC): human skin cancer tissue, mouse brain tissue, mouse kidney tissue, rat kidney tissue, mouse lung tissue, rat lung tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human p73.

Reactivity Notes

The supplier lists reactivity with human. UniProt places tumor protein p73 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Expressed in striatal neurons of patients with Huntington disease (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
