Product Overview
This rabbit polyclonal antibody recognizes protein kinase C and casein kinase substrate in neurons protein 1 (gene PACSIN1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 51.0 kDa, and the supplier reports an observed band at about 51 kDa.
PACSIN1 (syndapin I) is a neuron-enriched F-BAR protein that binds and bends membranes. It recruits dynamin and actin regulators such as N-WASP and cobl, supporting synaptic vesicle endocytosis and the growth and branching of neurites.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human glioblastoma tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): Jurkat cells, K562 cells.
Immunogen
Recombinant human PACSIN1 fragment (Gln76–Glu359), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 95.8% and 96.5%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places protein kinase C and casein kinase substrate in neurons protein 1 in the cytoplasm, cell projection, synapse and membrane. Tissue expression noted by UniProt (human): Highly expressed in brain and, at much lower levels, in heart and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
